TB-500 (Thymosin Beta-4)

TB-500 (Thymosin Beta-4) is a 43-amino acid naturally occurring peptide that is the most abundant member of the beta-thymosin family. Originally isolated from the thymus gland, Thymosin Beta-4 is now known to be expressed ubiquitously in virtually all cell types except red blood cells. Its primary intracellular function is sequestration of G-actin (monomeric actin), maintaining the pool of unpolymerized actin available for rapid cytoskeletal reorganization. This actin-regulating function makes Thymosin Beta-4 essential for cell migration, a process critical to wound healing, tissue remodeling, and inflammatory cell recruitment. Beyond actin regulation, Thymosin Beta-4 has been investigated for extracellular signaling activities including modulation of inflammatory pathways, metalloproteinase regulation, and promotion of angiogenesis. It has been studied in clinical trials for wound healing and cardiac repair applications.

TB-500 has been investigated for its dual intracellular and extracellular mechanisms. Intracellularly, Thymosin Beta-4 sequesters G-actin monomers in a 1:1 complex, preventing spontaneous polymerization and maintaining a dynamic actin pool available for rapid cytoskeletal reorganization during cell migration, division, and morphological changes. Researchers observed that this actin-buffering function is critical for lamellipodium formation and directional cell migration. Extracellularly, TB-500 has been investigated for anti-inflammatory signaling through NF-κB pathway modulation, reduction of pro-inflammatory cytokine expression, and promotion of anti-inflammatory macrophage polarization. Studies suggest that TB-500 also promotes angiogenesis through upregulation of VEGF and activation of endothelial cell migration. The peptide contains the active sequence Ac-SDKP (N-terminal tetrapeptide), which has been independently characterized as an anti-fibrotic factor that inhibits collagen deposition and promotes matrix remodeling. In cardiac models, researchers observed that Thymosin Beta-4 activated Akt/PKB survival signaling pathways.

Wound Healing

Malinda et al. (1999) investigated Thymosin Beta-4 in dermal wound models and observed accelerated wound closure, enhanced keratinocyte migration, and increased angiogenesis at wound sites [1].

Bock-Marquette et al. (2004) demonstrated that Thymosin Beta-4 activated integrin-linked kinase and Akt signaling in cardiomyocytes, promoting cell survival and migration in cardiac injury models [2].
